基因编码的光学探针有助于人们通过成像的手段实时、定量地研究复杂神经系统的信息整合(Feng et al.,2019; Jing et al.,2018; Jing et al.,2019; Patriarchi etal., 2018; Sun et al.,2018; Wang et al.,2018)。此前,我们将神经递质受体与荧光蛋白进行分子水平的融合和改造,开发出乙酰胆碱探针、多巴胺探针和去甲肾上腺素探针。现在,在发表的第一代多巴胺探针的基础上,我们开发出具有更高灵敏度的第二代探针,能够在活体果蝇中指示由“新奇感”(novelty)(Hattori et al., 2017)引起的单个多巴胺能神经元的多巴胺释放。
DA2m能够检测到活体单神经元的多巴胺释放
多巴胺神经元介导的在果蝇蘑菇体的可塑性被认为在新气味的感知中起重要作用 (Hattori et al.,2017)。利用我们新开发的DA2m绿色多巴胺探针,可以分别在熟悉气味和新型气味的情况下直接在体内对多巴胺动态进行成像。
我们最新开发的第二代、具有中等亲和力的多巴胺探针GRAB DA2m对饱和浓度的多巴胺具有>300%的荧光响应幅度。浓度依赖的荧光响应曲线指示EC50为90 nM。激发峰值、发射峰值与EGFP、GCaMP类似。
从下方视频可以看出,改良后的多巴胺探针DA2m有比已发表的DA1m (Sun et al.,2018) 更高的体内反应和更好的信噪比。
